Human Cultured NK Cells Electroporation Protocol & Parameters

Technical Overview & Cell State Insights

Electroporation parameters for primary purified natural killer (NK) cells and peripheral blood mononuclear cells (PBMCs) share similar characteristics. In their unstimulated, resting state, primary NK cells have a small diameter of approximately 7–8 μm, requiring significantly higher voltages than standard mammalian cell lines. Consequently, their transfection protocols can directly follow standard PBMC electroporation guidelines.

However, stimulated and cultured NK cells increase significantly in size during proliferation. Because larger cells require lower electric field strengths, cultured NK cells need much lower voltages than resting primary NK cells—comparable to standard suspension cell lines—and can be effectively transfected using the Cell Line Mode on Celetrix systems.

Optimization Highlights

Stimulation Methods: We evaluated various culture stimulation protocols and found that both feeder cell-co-culture and liquid-only media works. Feeder cell protocol may yield better results compared to liquid-only media.

Culture Duration: To achieve high transfection efficiency while maintaining optimal cell viability, NK cells are typically stimulated for 5-7 days prior to electroporation.

Key Variable: Because stimulation conditions directly impact cell size and viability, culture duration and electroporation voltage should be carefully optimized for your specific experimental workflow.

Performance Benchmark

NK cells stimulated for 6 days achieved an 80% GFP-positive rate at 48 hours post-electroporation with a GFP transposon, sustaining 50% positivity through 8 days of continued culture.
